Centre Joe Marchant has been left out of England's 25-man squad to face Wales at Twickenham on Saturday - with Manu Tuilagi set to start in the midfield.
As well as Tuilagi, flanker Courtney Lawes is on course to make his first appearance of this year's Championship after recovering from concussion.
But lock Joe Launchbury has been released back to Premiership side Wasps.
England head coach Eddie Jones will confirm his starting XV on Thursday.
Back row forward Sam Simmonds has recovered from a hip problem to be included in the 25 and will compete with Alex Dombrandt for a start at number 8.
Wing Jack Nowell has also been declared fit after recovering from concussion.
England started their campaign with a 20-17 defeat by Scotland at BT Murrayfield, before beating Italy 33-0 in Rome.
Saturday's game is their first at Twickenham this year, and attack coach Martin Gleeson says they must win to keep alive hopes of winning the Six Nations title.
"It's a big game and we're treating it like a quarter-final," Gleeson said.
"It's knockout stages for us. We know Wales fight like really hard and will make it really difficult for us.
"So we've got to go into this with a quarter-final mentality and go after Wales.
"If we don't get the result on Saturday, we're out."
